不同日龄白纹伊蚊雌蚊唾液腺总蛋白谱分析

摘要 目的:探讨不同日龄白纹伊蚊雌蚊唾液腺蛋白含量及总蛋白谱差异。方法:分别采用Pierce蛋白定量试剂盒和SDS-PAGE电泳方法,分析白纹伊蚊雌蚊各日龄唾液腺蛋白质定量及蛋白谱。结果:0日龄每对唾液腺蛋白质质量为(0.677±0.30)μg,3日龄为(1.454±0.46)μg,5日龄为(1.95±0.38)μg,7日龄为(1.733±0.46)μg,10日龄为(1.205±0.43)μg;SDS-PAGE检测到0日龄唾液腺总蛋白有12条主带,分别为15 kDa、18 kDa、30 kDa、32 kDa、34 kDa、40 kDa、45 kDa、48 kDa、50 kDa、62 kDa、68 kDa、>116 kDa 1条,还有其它比较弱的次带;3日龄以后的唾液腺总蛋白有12条带,但少了40 kDa条带,多了28 kDa条带。结论:不同日龄白纹伊蚊雌蚊唾液腺蛋白谱有一定差异。 Objective: To explore the difference of the salivary gland protein content and profile a- mong female Aedes albopictus at different age. Methods: The contents of salivary gland proteins were detected with Pierce protein assay kit, and protein profile was determined with SDS - PAGE electro- phoresis. Results: The amount of salivary gland proteins of 0 day, 3 days, 5 days, 7days and 10 days old female Aedes albopictus was 0. 677 ±0.30μg, 1. 454±0. 46 μg , 1.95 ±0. 38μg, 1. 733 ±0.46μ g and 1. 205±0.43μ g respectively. Twelve major bands of 15, 18, 30, 32,34,40, 45, 48, 50, 62, 68kDa and a band 〉 116 kDa of salivary gland proteins of 0 day old female mosquitoes were showed by SDS -PAGE electrophoresis. There were also 12 major bands for older female mosquitoes but with 28kDa instead of 40kDa. Conclusions: There are certain differences in the salivary gland profile a- mong female Aedes albopictus at different age.
