摘要
目的研究VEGF-C对继发性肢体淋巴水肿的治疗效果。方法手术和放射相结合的方法制作兔后肢淋巴水肿模型,之后将VEGF-C注射到局部水肿软组织中,对照组注射生理盐水。结果肢体水肿体积比较:治疗后同一时间两组比较均为P<0.05,差别有统计学意义;同组内不同时间比较,观察组均为P<0.05,差别有统计学意义;对照组均为P>0.05,差别无统计学意义。微淋巴管计数:治疗后同一时间两组比较,均为P<0.01,差别有明显统计学意义;同组内不同时间比较,均为P<0.01,差别有明显统计学意义。Flt-4、VEGF-C及vWF蛋白的变化:两组间比较,VEGF-C/β-actin,P<0.01,差别有显著统计学意义;FLT-4/β-actin,P<0.01,差别有显著统计学意义;vWF/β-actin,P>0.05,差别无统计学意义。结论重组VEGF-C蛋白局部注射后,水肿组织中VEGF-C、FLT-4等维持较高水平,促进淋巴管内皮细胞增生,加快淋巴回流网络的重建,可明显减少肢体水肿体积,并且随时间延长疗效逐渐增强,是治疗继发性肢体淋巴水肿的有效方法。
Objective: To study the curative effect on secondary limb lymphedema of human recombinant VEGF-C.Methods: Operation and radiation were combined to gain laboratory animal models.Human recombinant VEGF-C was injected into lymphedema limbs in the observation group but normal saline in the control group.Results: In the comparison of the swelling of lymphedema limbs,there was statistical significance(P﹤0.05)between the two groups at the same time,there was statistical significance(P﹤0.05)in the observation group among different periods but there was no significance(P〉0.05)in the control group.In the comparison of the micro-lymphatic vessel number,there was statistical significance(P﹤0.01)between two groups at the same period after treatment,and there was statistical significance(P﹤0.01)at different periods after treatment in the same group.Using Western blot to detect VEGF-C,flt-4 and vWF protein and compare between two groups,VEGF-C/β-actin was P0.05.Conclusion: VEGF-C injection can maintain high level of VEGF-C and flt-4,promote reconstruction of the lymphatic networks and lower the swelling of lymphedema limbs.It may be a good method to ease and cure the limb lymphedema.
