泰山地区回民糖耐量异常群体VDR受体基因多态性分析

摘要 目的探讨泰山地区回民群体糖耐量异常与维生素D受体基因多态性相关性。方法采用聚合酶链反应-限制性长度多态性技术,对回民糖耐量异常者和糖耐量正常回民检测VDR基因(Apa-I和Fok-I)多态性及等位基因分布频率,并检测VDR蛋白血液含量。结果糖耐量异常组Apal-I多态性AA、Aa和aa基因型分布率为10%、34.6%和55.4%,正常对照组相应为13.8%、42.5%和43.7%:糖耐量异常组Fok-I酶切多态性FF、Ff和ff基因型分布率为23.2%、41.1%和35.7%,正常对照组相应为13.8%、42.5%和43.7%;结论泰山地区回民群体糖耐量异常与维生素D受体基因多态性相关;VDR蛋白血液浓度和胰岛素抵抗在各型间显示差异性。 Objective: To investigate the vitamin D receptor gene polymorphism of Hui ethnic group with impaired glucose tolerance in Taishan region.Methods: The VDR gene(Fok,-I and Apa-I) polymorphism and allele distribution frequency were detected with polymerase chain reaction-restriction fragment length polymorphism(RFLP) method on sante Hui ethnic group with impaired glucose tolerance and normal Hui ethnic group,and also VDR protein in the blood of each group were detected.Results: In the impaired glucose tolerance group,VDR gene(Apal-I) polymorphism included AA(10%),Ff(34.4%) and ff(55.6%) three genotype,while they were 13%(AA),42.5%(Aa) and 43.7%(aa) in the normal control group;VDR gene(Fok-I) polymorphism included FF,Ff and ff genetype,the gene rate was 23.4(AA),41.1%(Aa) and 35.3%(aa) in impaired glucose tolerance group and 13.8%(AA),42.5%(Aa) and 43.7%(aa) in the normal control group.Conclusion: Hui ethnic group in Taishan region with impaired glucose tolerance is related to the vitamin D receptor gene polymorphism and VDR protein concentration in peripheral blood and insulin resistance is different significiantly among each genetype.
