摘要
目的:通过观察苏木乙酸乙酯提取物对大鼠同种异位心脏移植慢性排斥反应血清中TGF-β1水平的影响,为进一步探讨苏木乙酸乙酯提取物对慢性排斥反应的作用机制奠定基础。方法:以Wistar大鼠为供体,以SD大鼠为受体,Ono术式建立心脏移植模型。将上述大鼠术后24h给予35mg/kg/d环孢素A(CsA)灌胃,连续21天,以抑制急性排斥反应。21天后选择心脏跳动良好者随机分组。Ⅰ组为对照组,无药物治疗,灌服等体积的橄榄油;Ⅱ组为CsA组,按CsA12mg/kg/d灌服给药;Ⅲ组为苏木乙酸乙酯提取物组,按生药1.15g/kg/d灌服给药。给药7天后采集血液,用ELISA法检测血清中TGF-β1的含量。结果:与对照组相比,CsA组和苏木乙酸乙酯提取物组大鼠血清中TGF-β1的含量明显降低,差异具有统计学意义(P<0.01),苏木乙酸乙酯提取物组与CsA组相比,无显著差异(P>0.05)。结论:苏木乙酸乙酯提取物能降低心脏移植慢性排斥大鼠血清TGF-β1的含量,这可能是其抗慢性排斥反应作用的机制之一。
Objective: To observe the effect of ethyl acetate extracted from Sapanwood on the blood serum TGF - β1 of chronic rejection reactions after allogenic cardiac transplantation of rats, for the further study of sappan wood ethyl acetate extract on chronic rejection mechanism laying foundation. Methods: The allogenic cardiac transplantation model was established by using Wistar rats as donors and SD rats as receptors. After the suc- cess of the operation of continuous gavage for 21 days of 35mgc/kg/d CsA in rats, they were divided into three groups randomly. Group I was control group, rats in this group were dosed olive - oil; Group II was dosed 12mg/kg/d CsA; Group III was dosed ethyl acetate extracted from Chinese medicine sappan wood [ crudedrug 1.15g/kg/d]. After 7 days' dosing,the content of TGF- β1 in blood serum was tested by ELISA. Results: The content of TGF -β1 in Rats in Group I was notably higher than that in Group II and Group III (P 〈 0. 01 ), and the content of TGF - β1 in Rats in Group III was higher than that in Group II(P 〉0.05) . Con- clusion: The ethyl acetate extracted from Chinese medicine sappan wood can reduce cardiac allograft in rat se- rum TGF- β1 content,which may be one of the reasons against chronic reiection.
