摘要 本文用差速离心和凝胶层析法从大鼠肝、肾中分离纯化出GBA和GBN,分别比较二者与早孕绒毛GB的生化特性。结果指出:绒毛GB的电泳行为、pH曲线,K_n值及去垢剂影响等均与纯制GBA的相同或相近,但与GBN完全不同。由此证明早孕绒毛中的GB是GBA。本文还建立了绒毛GBA的荧光测定法。测定76例正常人流绒毛GBA活性值为263.7±65.4nmol 4MU·(mg protein·h)^(-1)。 Partially purified acid β-glucosidase (GBA) and neutral β-glucosidase (GBN) were obtained from rat liver and rat kidney using centrifugation and gel filtration chromatography. A comparative study showed that the β-glucosidase of chori-onic villi had the same or similar electrophoresis behavior, pH curve, Km value, detergents, effect, as GBA, but obviously different from GBN. Hence, It can be claimed that the β-glucosidase of chorionic villi wasGBA but not GBN. According to the results, a fluorimetry of chorionic villi GBA had been developed. The mean value of the GBA from 76 cases normal abortion was 263.7±65.4 nmol 4MU·(mg protein·2h)-1.
