摘要
目的研究透明质酸、透明质酸酶在垂体腺瘤侵袭性生物学行为中的意义。方法应用放射免疫法测定瘤组织中透明质酸,用酶联吸附测定法测定瘤组织中透明质酸酶活性及应用逆转录聚合酶链式反应(RT-PCR)测定瘤细胞中透明质酸酶基因表达。结果侵袭性垂体腺瘤患者瘤组织中透明质酸及透明质酸酶显著增高,透明质酸酶基因PH-20高表达,与非侵袭性垂体腺瘤组及正常脑组织组的差异有显著统计学意义(P<0.05)。结论透明质酸、透明质酸酶在垂体腺瘤侵袭性生物学行为中发挥作用,测定透明质酸和透明质酸酶对侵袭性垂体腺瘤患者诊断及预后判断有参考价值。
Objective To investigate the significance of hyaluronic acid and hyaluronidase on pituitary adenoma invasive biological behaviour. Methods Radioimmunity was utilized to determine the hyaluronic acid content of pituitary adenoma, enzyme linked sorbent assay was used to determine the hyaluronidase content of pituitary adenoma, reverse transcription-polymerse chain reaction was used to detect the gene expression of hyaluronidase. Results The contents of hyaluronic acid, hyaluronidase and the expression levels of hyaluronidase PH-20 gene in invasive pituitary adenoma were higher than these in no-invasive pituitary adenoma and normal brain tissure. Conclusions The hyaluronic acid and hyaluronidase have an important effect on pituitary adenoma invasive biological behaviour. It is a good value for invasive pituitary adenoma patients to determine the hyaluronic acid content and the hyaluronidase content.
