摘要
[目的]对细胞周期蛋白家族的序列进行生物信息学分析,从而研究物种间细胞周期蛋白的分类体系。[方法]选取已完成基因组测序的10个物种的细胞周期蛋白,按细胞周期蛋白中周期蛋白框结构域的序列完整性及结构域的数目进行分类,并进行多次筛选比对,分别构建具完整单周期蛋白框结构域和完整双周期蛋白框结构域细胞周期蛋白的系统发育树。[结果]随物种进化,细胞周期蛋白的序列类型和序列数量显著增加。对完整单域和完整双域细胞周期蛋白的聚类分析结果显示,原核生物的细胞周期蛋白、单细胞真核生物的PHO80与PCL、植物的P和U、动物的Y组成原核型细胞周期蛋白,并进化形成真核型细胞周期蛋白,真核型细胞周期蛋白的进化趋势是增加类型和数量,动物以增加细胞周期蛋白的类型为主,而植物则是以增加细胞周期蛋白类型中的成员为主;另一进化趋势是由单域向双域发展,物种进化程度越高,所具有的双域细胞周期蛋白的类型就越多。[结论]为物种间细胞周期蛋白的分子进化研究提供了参考。
[Objective] Bioinformatics was carried out on sequence of cyclin families.[Method] The cyclins of ten species which had accomplished genome sequencing,were classified according to the sequence integrity and number of domain.The phylogenetic trees with completed monocycle and dicycle domain were constructed respectively.[Result] The cyclin sequences from 10 model species were analyzed and the results suggested that numbers and types of cyclins in these species significantly increased during evolution.Phylogenetic analysis of the cyclins with single-complete cyclin box domain and doube-complete cyclin box domains indicated that prokaryotic cyclins were made up of cyclins of prokaryote,PHO80 and PCL in unicellular eukaryotes,P and U in plants and Y in animals,and from which eukaryotic cyclins were gradually evolved.The evolutionary trends of eukaryotic cyclins was increasing their numbers and types,and the evolutionary difference between animals and plants showed that animals mainly increased types of cyclins,whereas plants increased members of each cyclin type.Another evolutionary trends of eukaryotic cyclins was duplicating cyclin box domain,and the higher evolutionary of the species,the more types of cyclins with double cyclin box domain they had.[Conclusion] The study will provide reference for molecular evolution research of cyclin.
