摘要
[目的]比较南太湖地区不同区域荷叶中荷叶碱的含量。[方法]采用反相高效液相色谱法测定,以Agilent Eclipse XDB-C18柱(250 mm×4.6 mm,5μm)为固定相;以乙腈-水-三乙胺-冰醋酸(27∶70.6∶1.6∶0.78,V/V/V/V)为流动相;流速为1.0 ml/min;检测波长为270 nm;柱温为25℃。[结果]荷叶碱线性范围3.039~91.170μg/ml(R=1.000 0),平均回收率为98.48%,RSD为0.63%(n=6);7~8月采摘的荷叶中荷叶碱的含量最高(0.322%);在5个产地中,以德清县采摘的荷叶中荷叶碱的含量最高(0.465%)。[结论]荷叶碱含量随生长时间而变化;南太湖地区荷叶中荷叶碱含量差异不明显。
[Objective] To compare the alkaloid content in lotus leaf in southern Taihu Lake.[Method] The RP-HPLC method was adopted,with Agilent Eclipse XDB-C18 column(250 mm× 4.6 mm,5 μm) as the stationary phase,Acetonitrile-water-triethylamine-aceticacidas(27∶70.6 ∶1.6∶0.78) as mobile phase,flow rate 1.0 ml/min,detection wavelength 270 nm,column temperature 25 ℃.[Results] Lotus leaf alkaloid was linear in the range of 3.039-91.170 μg/ml(R=1.000 0),the average recovery rate was 98.48%,RSD= 0.63(n=6).The alkaloid content was highest(0.322%) in lotus leaf picked during July-August;among five producing area,Deqing County was the highest(0.465%).[Conclusion] The alkaloid content in lotus leaf varied with growth time,there is no significant difference in southern Taihu Lake.
