摘要
目的探讨肌醇依赖性激酶1α(IRE1α)对非洲爪蟾心脏发育的影响。方法通过显微注射IRE1αmRNA实现基因过表达,注射IRE1α或其下游基因XBP1特异性反义寡核苷酸(MO)实现基因封闭;利用全胚胎原位杂交方法检测基因表达,免疫组化方法检测体节形成。结果过表达IRE1α对心脏发育无明显影响;利用MO封闭IRE1α或XBP1后,心脏明显减小,形态异常,心脏发育相关基因nkx2.5表达下降,但对体节无影响。结论 IRE1α影响非洲爪蟾心脏的发育,可能是通过XBP1发挥作用。
Objective To investigate the heart development in Xenopus lavies. Methods effects of inositol-requiring enzyme-1α(IRE1α) on the The mRNA was microinjected to overexpress IRE1α and the gene-specific antisence oligonucleotids morpholino (MO) was microinjected to knockdown IRE1α and XBP1. The whole mount in situ hybridization was used to detect the gene expression of nkx2. 5. The somite development was measured by immunohischemistry. Results The overexpressed IRE1α did not significantly affect the heart development in Xenopus lavies, while the heart was seriously diminished, the gene expression of nkx2. 5 was decreased, but the,somite was not obviously changed in the IRE1α or XBP1 knockdown embryos. Conclusion IRE1α plays an important role in the heart development probably in an XBP1-dependent pathway.
