钙蛋白酶在大鼠心肌肥厚中的作用及调节 被引量:1

Function and regulation of calpain in rat with myocardial hypertrophy

摘要 目的探讨压力超负荷下,钙蛋白酶参与心肌肥厚的信号转导机制。方法16只SD大鼠随机均分为模型组和对照组。模型组采用不完全结扎腹主动脉法建立大鼠高血压心肌肥厚模型。放射免疫法检测两组心肌心房肽(ANF)、血管紧张素Ⅱ(AngⅡ)水平;Western blot法检测左室心肌钙蛋白酶的表达水平。结果模型组术后血压、心肌ANF、AngⅡ含量、左室心肌钙蛋白酶同工酶u表达水平明显高于对照组(P<0.01)。结论心肌肥厚组织中钙蛋白酶同工酶u表达增加,钙调神经磷酸酶信号通路参与了心肌肥厚的发病过程。 Objective To investigate the signal transduction mechanism of calpain in the myocardial hypertrophy under overloaded pressure. Methods Sixteen SD rats were equally randomized into groups of A(hypertensive myocardial hypertrophy model by incomplete constriction of abdominal aorta) and B(sham operation as control). The levels of atrial natriuretic factor(ANF) and angiotensin Ⅱ (Ang Ⅱ) in myocardium were detected using radioimmunoassay and the calpain expression in left ventricular myocardium was detected by Western blot in two groups. Results Blood pressure,contents of ANF and Ang Ⅱ in myocardium and the level of u-calpain expression in left ventricular myocardium were higher in group A than those in group B after operation(P〈0. 01). Conclusion The expression of u-calpain is increased in hypertrophic myocardium. Calcineurin signaling pathway involves in the pathogenesis of myocardial hypertrophy.
