N-乙酰半胱氨酸对重症急性胰腺炎大鼠血-脑屏障及脑组织损害的保护作用 被引量:1

摘要 目的探讨N-乙酰半胱氨酸(NAC)对重症急性胰腺炎(SAP)大鼠血-脑屏障(BBB)及脑组织损害的保护作用。方法 30只SD大鼠随机分为假手术组(SO组)、SAP组、NAC干预组(NAC组),每组10只大鼠。从胰胆管逆行注入5%牛磺酸钠(0.1 ml/100 g)制作SAP模型。分别于造模后3 h、6 h、12 h将大鼠处死,取脑。采用光镜、电镜观察大鼠脑组织病理变化,免疫组化染色检测与BBB有关的3种细胞骨架蛋白:胶质纤维酸性蛋白(GFAP)、凋亡相关基因(Bcl-2)和基质金属蛋白酶-9(MMP-9)的表达。结果 NAC组各时间点大鼠脑组织病理评分明显低于SAP组(均P<0.01)。SAP组和NAC组各时间点Bcl-2、GFAP、MMP-9阳性细胞数均明显多于SO组(均P<0.01)。NAC组各时间点Bcl-2、GFAP阳性细胞数均明显多于SAP组(均P<0.01),MMP-9阳性细胞数明显少于SAP组(均P<0.01)。结论 NAC能调节SAP大鼠的炎症因子表达,明显减轻其BBB的损害,对脑组织具有保护作用。 Objective To study the protective effects of N-acetylcysteine (NAC) on blood-brain barrier (BBB) and brain tissues injure in rats with severe acute pancreatitis (SAP). Methods Thirty SD rats were randomly divided into sham operation group (SO group), SAP group and NAC intervention group (NAC group), with 10 rats in each group. SAP model was induced by retrograde injection of 5% sodium taurocholate (0.1 ml/100 g) into the bilipancreatic duct. All the animals were sacrificed at 3 h, 6 h, 12 h after induction of pancreatitis. Histopathological changes in rats brain tissue were detected with light microscope and electron microscope. Immunohistochemistry was detected expression of tree cytoskeletal protein about BBB which is glial fabrilary acidic protein( GFAP), Bcl-2 and Matrix Metalloproteinase-9 (MMP-9). Results The pathology scores in NAC group at each time points were significantly lower than that in SAP group ( all P 〈 0.01 ) i Positive cells of Bcl-2, GFAP and MMP-9 in the SAP and NAC group at each time points were significantly more than those in SO group ( all P 〈 0. 01 ). The positive cells of Bcl-2 and GFAP in NAC group at each time points were significantly more than those in SAP group (all P 〈 0.01 ). The positive cells of MMP-9 in NAC each subgroup were significantly less than those in SAP group ( all P 〈 0. 01 ). Conclusion NAC can regulate the expresstion of inflammation factors, significantly reduce the blood-brain barrier damage and have a protective effects on brain tissues in rats with SAP.
