摘要
旨在研究二甲双胍(Metformin)对PRKAG3基因表达量以及对猪生长性能及肉质的影响。挑选80kg左右的杜洛克×长白×大约克三元杂交(DLY)阉公猪10头,随机分为对照组和试验组。试验组日粮中添加400mg·kg-1体质量的Metformin,对照组日粮除不含Metformin外,其它日粮成分与试验组相同。2周试验结束后所有猪只全部屠宰,测定生长性能、AMPK活性、PRKAG3基因表达量、AMPKγ3蛋白表达量、胴体品质和肉质性状。结果表明:1)日粮中添加400mg·kg-1体质量的Metformin后,试验组AMPK活性、PRKAG3基因mRNA含量和AMPKγ3蛋白表达量分别比对照组提高了13.92%、4.39倍和35.53%(P<0.05)。2)试验组采食量和平均日增体质量分别比对照组下降13.80%(P<0.05)和13.14%,而料肉比则比对照组提高6.77%。3)试验组胴体长显著低于对照组(P<0.05),屠宰率、背膘厚、眼肌面积和瘦肉率均低于对照组,但差异均不显著(P>0.05)。4)试验组pH2、滴水损失、a值低于对照组(P>0.05);pH1、剪切力和熟肉率高于对照组(P>0.05);L值和b值则极显著高于对照组(P<0.01)。结果提示,Metformin可激活PRKAG3基因表达,进而可能影响猪生长性能和肉质。
This study was conducted to investigate the effect of Metformin on PRKAG3 gene expression, growth performance and pork quality in pigs. 10 DLY pigs (80 kg) were randomly divided into control and experimental groups, pigs in experimental group fed diet supplemented with Metformin at dose of 400 mg · kg^-1 body weight. After 2 weeks, all pigs were slaughtered to determine growth performance, AMPK activity, PRKAG3 gene expression, AMPK73 protein levels and pork quality traits. The results showed that, compared with control group, experimen- tal group had 13.92% higher AMPK activity (P〈0.05), 4.39-fold PRKAG3 gene expression (P〈 0.05) and 35. 53% higher AMPK73 protein level (P〈0.05). Pigs in experimental group had 13.80% lower feed intake (P〈0.05), 13.14% lower average daily gain and 6.67% higher F/G than that in control group. Experimental group reduced dressing percentage, lean percentage, eye lean area, and backfat thickness(P〉0.05), and significantly reduced carcass length(P〈 0.05). Compared with control group, experimental group had lower pH2, drip loss, a values, higher pHi, shear force and cooked meat percentage, but there was no significant difference between control and exp L and b values (P〈0. erimental groups(P〉0.05).
