热休克蛋白70对大鼠牙髓细胞高温损伤的影响 被引量:3

摘要 目的:研究预热处理诱导的热休克蛋白70(HSP70)对受高温损伤的大鼠牙髓细胞的影响。方法:将体外培养的大鼠牙髓细胞随机分为4组,分别为正常对照组(C组):37℃常规培养18h;预热处理组(pH组):37℃常规培养9h后42℃培养1h,再复温培养8h;高温损伤组(SH组):37℃常规培养9h后46℃培养1h,再复温培养8h;预热处理+高温损伤组(pH+SH组):42℃培养1h后恢复常规培养8h,再置于46℃培养1h后恢复常规培养8h。以Western Blot检测细胞内HSP70的表达变化;以四唑盐(MTT)比色法、吖啶橙/溴化乙啶(AO/EB)双染法、细胞形态学观察评价细胞存活能力。结果:Western Blot显示,与C组相比较PH组HSP70表达上调;MTT比色法、AO/EB双染法、细胞形态学观察均显示pH+SH组较SH组的细胞存活能力强。结论:预热处理可诱导大鼠牙髓细胞中的HSP70表达上调,减轻高温对牙髓细胞的损伤。 Objective: To investigate the effects of heat shock protein 70(HSP70) induced by preheating treatment on lethal heat-induced injury to rat pulp cells.Methods: Pulp cells were randomly divided into four groups as following: I control(C)group;II preheating(PH) group was subjected to sublethal heat(42℃×1h);III severity hyperthermia(SH) group was subjected to lethal heat(46℃×1h);IV preheating + severity hyperthermia(pH+SH) group was treated with sublethal heat(42℃×1h) 8h before subjected to lethal heat(46℃×1h).The expression of HSP70 was detected by Western Blot;the cell viability was measured by MTT,AO/EB and cytomorphology.Results: As Western Blot revealed: compared with C group,the expression of HSP70 in PH group was upregulated;as concluded respectively by MTT,AO/EB and cytomorphology,the cell in PH+SH group was of great viability.Conclusion: Preheating treatment can upregulate the expression of HSP70 in rat pulp cells,thus protecting the cells from lethalheat injury.
