摘要
目的比较P-糖蛋白(P-gp)与谷胱甘肽S转移酶π(GST-π)在鼻咽癌细胞TW03及其多药耐药细胞TW03/DDP中的表达差异,进一步探讨鼻咽癌多药耐药的机制。方法制备鼻咽癌细胞TW03及其多药耐药细胞TW03/DDP的细胞爬片,利用免疫细胞化学技术,检测P-gp与GST-π在两种细胞中的表达情况。结果 GST-π在TW03/DDP中表达明显高于TW03(P<0.01),而P-gp在TW03/DDP中表达明显低于TW03(P<0.01)。结论 GST-π可能是TW03/DDP导致多药耐药中的重要原因之一,P-gp降低的原因需要进一步研究。
Objective To compare the expression differences of P- glucoprotein (P- gp) and glutathione S-transferase 7r (GST- π) between nasopharyngeal cancer cells TW03 and its multi- drug resistance ceils TW03/DDP, zncl to further studv the multi - dru~ resistance mechanism of nasot3harvnzeal cancer. Methods The cell slide preparation for nasopharyngeal cancer cells TW03 and its multi- drug resistance cells TW03/DDP was performed. An immunocytochemistry technique was used to detect the expression of P - gp and GST- rt in the two mentionedabove cells. Results The expression of GST - πin TW03/DDP was statistically higher than that in TW03 ( P 〈0.01), while the expression of P - gp in TW03/DDP was statistically lower than that in TW03 ( P 〈 0.01). Con- clusion GST - zt may play an important role in TW03/DDP - induced multi - drug resistance. The mechanism ofdecreased P - gp needs further study.
