摘要
目的研究鸡骨草醇提物对四氯化碳所致大鼠肝纤维化的影响。方法复制肝纤维化大鼠模型,随机分成5组:模型组,阳性组(秋水仙碱0.2mg/kg),鸡骨草醇提物低、中、高剂量组(20、40、80mg/kg),并设立正常组。连续灌胃给药30天。检测大鼠血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)水平和谷胱甘肽-过氧化物酶(GSH-Px)活性以及肝组织中羟脯氨酸(Hyp)的表达,采用Masson染色观察肝组织纤维化程度。结果与模型组比较,鸡骨草醇提物组显著降低肝纤维化大鼠血清中AST、ALT水平,提高GSH-Px活力(P<0.05),同时减少肝组织中Hyp含量(P<0.05),并减少大鼠肝脏胶原纤维。结论鸡骨草醇提物对CCl4所致的肝纤维化大鼠具有保护作用。
Objective To study the effect of abrus cantoniensis alcohol extract on carbon tetrachloride (CC14) induced liver fibrosis in rats. Methods Rat liver fibrosis models were established, all of them were randomly divid-ed into five groups: model group, colchicine group ~,0.2 mg/kg), low - , medmm - and high -- close groups ot Abrus cantoniensis alcohol extract (20,40, 80 mg/kg), as well as normal group were set up, drug intragastric ad- ministration durated for 30 days. The blood was collected and the levels of alanine aminotransferase (ALT), aspar- rate aminotransferase (AST) and GSH- Px in serum were examined, while the content of Hyp in liver tissue was tested, and the severity of liver fibrosis was observed by Masson- staining. Results Compared to model group, abrus cantoniensis extract could significantly decrease the levels of AST and ALT in injured liver tissues induced by CC14, while enhanced GSH-Px activity ( P 〈0.05), as well as reduced the content of Hyp in liver tissues ( P 〈 0.05). and liver fibrosis was alleviated. Conclusion The results indicated that abrus cantoniensis alcohol extracthas protective effect on CCLt induced liver fibrosis in rats.
