摘要
目的 探讨IL-8在脑缺血损伤中的变化。方法 采用改良Zea Longa线栓法大鼠MCAO模型,将 大脑中动脉(MCA)闭塞不同的时间。应用双抗体夹心间接ELISA法检测缺血组和对照组大鼠受伤脑组织及血清 中IL-8浓度。结果 闭塞3h缺血侧半球湿重大于非缺血侧(P<0.05),并维持至48h;缺血组脑组织IL-8含量在 MCA闭塞3h处于较低水平,随后逐渐升高,至6h达高峰,随后又降低;血清中IL-8含量在缺血3h达高峰,随后缓 慢下降。空白组、假手术组脑组织内IL-8含量高于缺血组(P<0.01),同一大脑非损伤侧高于损伤侧。结论IL-8 具有双重作用,既参与脑组织的正常代谢及生理功能,又参与了脑缺血损伤的病理生理过程。
Objective To study the change of the level of interleukin-8(IL-8) in cerebral ischemia injury. Methods The middle cerebral artery (MCA) was occluded for different time with modified Zea Longa's MCAO model. The level of IL-8 in injured brain tissue and blood serum was detected with double-antibody sandwich enzyme-linked immunosorbent assay,both in ischemia group and in control group. Results The weight of the injured brain hemisphere is heavier than that of uninjured hemisphere from 3 hours to 48 hours after cerebral ischemia(P<0.05). The content of IL-8 in injured brain tissue was in lower level at 3 hours after ischemia,and then increased,peaked at 6 hours,and then decreased. The concentration of IL-8 in serum peaked at 3 hours after ischemia,and then decreased slowly. The content of IL-8 in brain tissue of control group was higher than that in ischemia group (PRO.01). Conclusion IL-8 has double roles it play a role in metabolism and function in normal brain tissue and a role in pathophysiological course of cerebral ischemia injury.
