HPLC法测定连菊清热胶囊中连翘苷的含量

Determination of contents of phillyrin in Lianjuqingre capsule by HPLC

目的建立HPLC法测定连菊清热胶囊中连翘苷含量的方法。方法采用HPLC法,选用C_(18)柱(4.6 mm×250mm,5μm),选用乙腈-水(21:79)为流动相,流速1.0 ml/min,检测波长230 nm,柱温25℃。采用校正曲线法测定连翘苷含量。结果连翘苷的线性范围为0.424-2.120μg;回归方程为y=948.48x+149.17,r=0.999 2,平均回收率为97.85%,RSD为2.44%(n=6)。结论该HPLC法测定样品灵敏度高,重复性好,能有效地控制连菊清热胶囊的质量。

Objective To establish a HPLC method for determining the content of phillyrin in Lianjuqingre capsule.Methods C_（18） column（250 mm×4.6 mm,5μm）was applied with the mobile phase of acetonitrile-water（21：79）at 25℃and the flow rate was 1.0 ml/min.The detective wavelength was set at 230 nm.The content of phillyrin in Lianjuqingre capsule was determined by HPLC method.The calibration curve method was investigated to determine the content of phillyrin.Results Phillyrin showed a good linear relationship between the input volume and peak area in the range of 0.424-2.120μg,and the equation was y=948.48x ＋ 149.17. The average recovery was 97.85%and RSD was 2.44%（n=6）.Conclusion The established method is sensitive and repeatable, and can effectively control the quality of Lianjuqingre capsule.
