中药益脂康降脂降糖作用的实验研究

Experimental Study of Effect of Chinese Medicine Yizhikang Lipid-lowering Hypoglycemic

目的研究益脂康对2型糖尿病合并高脂血症的降糖降脂作用及与盐酸二甲双胍+非诺贝特的对比。方法采用高脂饲料联合琏脲佐菌素(STZ)30mg/kg腹腔注射,建立2型糖尿病合并高脂血症大鼠模型,查空腹血糖(FBG)、总胆固醇(TC)、甘油三脂(TG)、谷丙转氨酶(ALT)、肌酐(Cr)。将造模成功的33只大鼠,按血糖高低随机分为3组:中药组(益脂康,0.15mg/kg)、西药组(盐酸二甲双胍60mg/kg+非诺贝特24mg/kg)及模型组,前2组按设计方案灌胃治疗,1次/d,连续给药1个月、2个月后分别复查FBG、TC、TG、ALT、Cr,并取未作任何处理的正常组、未予药物治疗的模型组及用药1个月、2个月后西药组、中药组的组织进行HE染色(肝、胰腺、肌肉、肾脏)、免疫组化(肝、胰腺、肌肉)及超微病理。正常组给予正常饲料,模型组给予高脂饲料。结果益脂康可以显著降低FBG、TC及TG,且对肝脏及肾脏功能的损害较小。结论益脂康对2型糖尿病合并高脂血症,能促进胰岛β细胞损伤恢复、增加胰岛素分泌及受体数目,对提高胰岛素敏感性,降低胰岛素抵抗等有作用。

Objective Purpose study on Yizhikang to hypoglycemic lipid-lowering effects of type 2 diabetes with hyperlipidemia and contrast to Metformin Hydrochloride ＋ fenofibrate.MethodUsing high fat diet and streptozotocin 30mg/kg intraperitoneal injection,building type 2 diabetic patients with hyperlipidemia rat model,and Searching fasting blood,glucosetotal cholestero,triglyceride,alanine aminotransferase,blood creatinine.Following the fasting blood glucose,take the 33 rat model into three groups：Chinese medicine（yizhikang,0.15mg/kg）,Western medicine（Metformin hydrochloride 60mg/kg ＋ fenofibrate 24mg/kg）,Model group,The first two groups give consecutive treatment,1 time / day,continuous administration of one and two months latter,reviewing FBG、TC、TG、ALT、Cr,and stained by HE,immunohistochemical and ultrastructural pathology of three groups,giving the normal group with normal diet,model group with high fat diet.ResultYizhikang can significant reduction in FBG,TC,TG,and having less damaging of the liver and kidney function.Conclusion Yizhikang can promote islet β cell injury recovery,increase insulin secretion and receptor number,improve insulin sensitivity,reduce insulin resistance.
