摘要
目的 研究反义癌基因与抑癌基因联合抑制肝癌细胞生长的效果。方法 利用基因重组方法构建携带 p16基因、p5 3基因的融合表达载体 (pcDNA16 5 3) ,利用基因合成仪体外合成N ras、c myc反义寡核苷酸 ,并将N ras、c myc反义寡核苷酸与pcDNA16 5 3融合表达载体转染到人肝癌细胞系SMMC772 1细胞中 ,观察肝癌细胞生长情况及软琼脂集落形成能力。结果 联合治疗组肝癌细胞增殖速度最慢 ,软琼脂集落形成个数最少 ,只有 2个。结论 利用相关基因联合治疗效果明显优于利用个别基因的治疗效果。
Objective\ To study the cooperative repressive effect of antisense oncogene and cancer suppressor gene on human heptic carcinoma cell. Methods\ Fusion expression victor of pcDNA16-53 and antisense oligonucleotide of N-ras and C-myc was transfected to human heptic carcinoma cell line SMMC7721,and the growth of heptic carcinoma cell and ability of forming colonies on soft agar were observed. Results\ The slowest growth of heptic carcinoma cell and the lest colonies formed on soft agar of the combination therapy group were found. Conclusion\ The therapeutic effect of related gene combination therapy is obviously better than that of singular gene therapy.\;
