MMP-9及VEGF蛋白在胶质瘤中的表达及其临床意义

摘要 目的观察基质金属蛋白9(matrix metalloproteinase 9M MP-9)及血管内皮生长因子(vascular growth factor VEGF)蛋白在脑胶质瘤中的表达,并探讨其临床病理意义。方法利用免疫组织化学检测技术,检测63例胶质瘤组织中MMP-9及VEGF蛋白的表达,并分析其表达与病理分级之间的关系。结果①MMP-9在胶质瘤中阳性表达为76.19%,MMP-9蛋白阳性产物主要分布在胶质瘤组织的胞浆及细胞间质;低分化瘤MMP-9蛋白表达阳性率(87.87%)显著高于高分化(63.33%)(P=0.037)。②VEGF蛋白在胶质瘤中阳性表达为74.76%,VEGF蛋白阳性产物主要分布于胶质瘤组织的血管内皮细胞;低分化者VEGF蛋白表达阳性率(90.90%)显著高于低分化者(56.67%)(P=0.003)。③MMP-9及VEGF蛋白表达呈正相关。结论 MMP-9及VEGF蛋白可作为揭示胶质瘤恶性程度的指标,并在胶质瘤的浸润转移过程中发挥重要作用。 Objective To obserrs protein expression of matrix metalloproteinase 9 (MMP-9) and vas- cular endothelial growth factor (VEGF) was studied in human glioma patients in relation to clinicopathological characteristics. Methods The expression of MMP-9 and VEGF in 56 human glioma tissues were examined by immunohistochemically method and analyzed the relationship between the protein expression and the pathological grades. Results (1)The expression rate of MMP-9 was 76. 19% in human glioma tissue. Immunostaining of MMP-9 expressed in the cytoplasm and intercellular substance of glioma. The expression of MMP-9 in the poorly differentiated glioma (87. 87% ) was higher significantly than in well-differentiated glioma (63. 33% )( P = O. 037). (2)The expression rate of VEGF was 74.76% in human glioma tissue. Immunostaining of VEGF ex- pressed in vascular endothelial ceils of glioma, The expression of VEGF in the poorly differentiated glioma (90. 90% ) was higher significantly than in the well-differentiated glioma (56. 67% ) (P = 0. 003 ). (3)There was a positive cor- relation between the expression of MMP-9 and VEGF. Conclusion MMP-9 and VEGF may play an important role in the invasive and metastatic progression of human glioma. It suggests that the detection of MMP-9 and VEGF expression in tumor tissues may be a new biological markers in malignant degree of human glioma.
