黄连解毒组方颗粒剂对小鼠肠道菌群的影响

Effect of Huanglian Jiedu Prescription Granule on Mice Intestinal Microflora

目的:探讨黄连、黄芩、黄柏和栀子等四种单味中药颗粒剂按传统黄连解毒汤配方制成的黄连解毒组方颗粒剂对小鼠肠道菌群的影响。方法:选用40只昆明种小鼠,随机分成4组(正常对照组和高、中、低剂量组),每组10只;正常对照组只灌胃生理盐水,高、中、低剂组量用不同剂量的黄连解毒颗粒剂组方灌胃,观察各组小鼠饮食、运动、体重等情况,灌胃14天后,对各组小鼠的淋巴细胞数、细菌移位现象、肠道菌群和肠道病理切片等进行分析。结果:黄连解毒组方颗粒剂高、中剂量组小鼠在试验期间体重增长缓慢,灌胃14天后其肠道益生菌较正常对照组明显减少,差异有统计学意义(P<0.05);球杆菌比大于1/3,出现了菌群结构失调;回肠黏膜变薄,绒毛显著萎缩变短脱落,细菌移位频率(%)与正常组比较差异明显(P<0.05),淋巴细胞数明显少(P<0.05)。低剂量组体重增加明显,肠道菌群比例正常,淋巴细胞数未见变化,无细菌移位现象,回肠黏膜和盲肠均正常。双歧杆菌和乳酸杆菌等益生菌数量增加,条件致病性肠球菌和大肠埃希菌较正常组减少。结论:高、中剂量的黄连解毒组方颗粒剂在用药一段时间后,对肠道益生菌有抑制作用及对肠道粘膜有损伤作用。低剂量使用时有调节肠道菌群平衡的作用。

Objective： To investigate effects of Huanglian Jiedu prescription granules to the balance of mice intestinal flora ,which gran- ules be made of four single traditional Chinese medicines named Coptis,Huang Qin,cortex phellodendri and gardenia according the tradi- tional way. Methods ：40 Kunming mice were randomly divided into 4 groups （high- middle-and low-dosage groups and normal control group）, each of which containsl0 mice. the normal control group were imbued with saline water everyday ,and other groups with different doses of Huanglian Jiedu prescription granules. And then, changes of diets, weights and behaviors of the mice were observed after 14 days gastric lavage, and Lymphocyte count, Bacterial translocation, Intestinal flora and pathological section of ileum of the mice were- investigated. Results： Mice with Huanglian Jiedu prescription granules in high-, middle-dosage groups got slow weigtlt growths during the test; gavage after 14 days, intestinal probiotics of the mice decreased significantly than the normal control group, the difference of which was statistically significant （ P 〈 0.05 ）; Ball coli came to more than 1/3, the microflora flora balance was destroyed; ileal mucosa became thinner, villous were significantly atrophied and fell off, the bacterial translocation frequency （ % ） compared with the normal group differences was significant （P〈0.05）, decrease of the lympilocyte count was significant （ P〈0.05 ）. The low-dosage group gained weight Significantly, Intestinal flora proportion and lymphocyte count kept in normal station, no bacterial translocation, ileal mucosa and cecum were normal. Bifidobacterium and Lactobacillus probiotics increased, conditional pathogenic Enterococcus and Escherichia coli decreased compared with the normal group.Conclusion ：Administration of Huanglian Jiedu prescription granules in high- and mid- dle-dosage after a period of time, there were inhibitory effects to intestinal probiotics and some injury to the intestinal mucosa.On the oth- er hand, the low-dosage Administration can benefit the balance of intestinal flora.
