吸入糖皮质激素对哮喘大鼠气道重塑的影响

摘要 目的研究糖皮质激素对气道重塑指标α-平滑肌动蛋白(α-SMA)和I型胶原的影响。方法将大鼠随机分为正常对照组、OVA组即哮喘模型组和布地奈德组各10只。采用免疫组化法测定肺组织中α-SMA的表达水平;酶联免疫吸附法测定成纤维细胞提取物中I型胶原的表达强度;并研究布地奈德(BUD)干预治疗后气道重塑指标α-SMA和I型胶原的表达水平。结果①OVA组肺中支气管的α-SMA的表达较正常对照组增强(P<0.05);布地奈德组肺中支气管的α-SMA的表达较OVA组减弱(P<0.05)。②OVA组的成纤维细胞(LF)提取物中的I型胶原含量高于正常对照组(P<0.05);布地奈德组肺中支气管的I型胶原含量低于OVA组(P<0.05)。结论布地奈德降低α-SMA和I型胶原的表达,能抑制气道重塑的发生。 Objective :To study the effects of glucocorticoid on type I collagen and or-smooth muscle actin (α-SMA) in asthma murine model. Methods : Pathogen-free 30 rats were divided into the 3 groups : blank group, OVA group and BUD group, α-SMA and type I collagen level in lung fibroblast were measured by immunohistochemistry and ELISA respectively. Meanwhile we examined the effects of inhaled glucocorticoid on the α- smooth muscle actin (α-SMA) of pulmonary branchi and type 1 collagen in the lung fibroblast (LF) which were the main signature index of airway remodeling. Results: ①The α-SMA expression in OVA group was more enhanced than those in blank group ( P 〈 0.05), and it in BUD group was more attenuated than those in OVA group (P 〈 0.05). ② The expression of the type I collagen in LF in OVA group was more enhanced than those blank group, and it in BUD group was more attenuated than those in OVA group ( P 〈 0.05 ). Conclu- sion: Budesonide can downregulate the expression of the α-SMA and type I collagen, so the treatment with Budesonide can inhibit the airway remodeling.
