大连市肠道病毒71型基因特征分析

Genetic characterization of Enterovirus 71 in DaLian City

摘要 目的了解大连市手足口病病原体肠道病毒71型(EV71)的VP1区段的基因特征。方法用RT-PCR方法从手足口病患者临床标本中扩增EV71的VP1基因全序列,利用软件进行同源性分析和构建系统发生树。结果测序获得的9株病毒基因与C4亚型代表株具有较高的核苷酸和氨基酸序列同源性。在系统进化树上,9株病毒基因与C4基因亚型代表株处于同一分支。结论大连市EV71病毒株属C4亚型。 Objective To explore the molecular characteristics of the VP1 coding fragment of enterovirus 71（EV71） strains isolated from Dalian.Method The complete VP1 coding gene of EV71 strains amplified by RT-PCR from clinical specimens of children with hand-foot-and-mouth disease（HFMD）in Dalian were sequenced and analyzed by software.Result The analysis indicated that the nucleotide and amino acid homogeneity of 9 strains were higher with C4 representative isolate and clustered with reference strains of C4 subgenotype in the phylogenetic tree.Conlusion The EV71 isolated strains caused HFMD in Dalian belonged to subgenotype C4.
