摘要
目的探讨肿瘤转移抑制基因KISS-1及基质金属蛋白酶-9(MMP-9)与胃癌侵袭、转移的关系,为研究胃癌的转移机制及临床治疗提供理论基础。方法采用免疫组织化学法检测36例胃癌组织及36例正常胃黏膜组织中KISS-1及MMP-9蛋白的表达情况,分析其与胃癌患者各临床病理指标的关系及二者表达的相关性;采用Western blot法检测不同侵袭能力胃癌细胞株BGC-823(高侵袭)和MKN-28(低侵袭)中KISS-1和MMP-9蛋白的表达。结果KISS-1蛋白在胃癌组织中的阳性表达率显著低于正常胃黏膜组织(P<0.05),并且其低表达与淋巴结转移密切相关(P<0.05);MMP-9蛋白在胃癌组织中的阳性表达率显著高于正常胃黏膜组织(P<0.05),并且其高表达与胃癌的浸润深度和淋巴结转移均密切相关(P<0.05);KISS-1与MMP-9蛋白的表达呈负相关(P<0.05)。KISS-1蛋白在高侵袭胃癌细胞株中的表达低于低侵袭细胞株,但差异无统计学意义(P>0.05);MMP-9蛋白在高侵袭胃癌细胞株中的表达高于低侵袭细胞株,差异有统计学意义(P<0.05)。结论 KISS-1蛋白的低表达和MMP-9蛋白的高表达可能与胃癌的浸润、转移有关,二者有望成为判定胃癌侵袭和转移能力的指标。
Objective To explore relationship between metastasis suppressor gene KISS-1,matrix metalloproteinase-9(MMP-9) and the invasion,metastasis of gastric carcinoma,in order to provide a theoretical foundation for investigation of metastatic mechanisms and clinical treatment of gastric carcinoma.Methods The expressions of KISS-1 and MMP-9 protein in 36 cases of gastric carcinoma and 36 cases of normal gastric epithelia were detected by immunohistochemistry,and the relationships between them and clinical pathologic parameters were analyzed.The expressions of KISS-1 and MMP-9 protein in BGC-823 cell lines with high invasive ability and MKN-28 cell lines with low invasive ability were detected by Western blot.Results The positive expression rate of KISS-1 protein in gastric carcinoma was significantly lower than that in normal gastric epithelia(P0.05),and the low expression of KISS-1 protein in gastric carcinoma was correlated with lymph node metastasis(P0.05).The positive expression rate of MMP-9 protein in gastric carcinoma was significantly higher than that in normal gastric epithelia(P0.05),and the high expression of MMP-9 protein in gastric carcinoma was correlated with the depth of invasion and the metastasis of lymph node(P0.05).The expression of KISS-1 has negative correlation with the expression of MMP-9(P0.05).The protein level of KISS-1 was slightly lower in BGC-823 cells than in MKN-28 cells,but the difference was not statistically significant(P0.05).The protein level of MMP-9 was significantly higher in BGC-823 cells than that in MKN-28 cells(P0.05).Conclusion Low expression of KISS-1 and high expression of MMP-9 protein may be related with invasion and metastasis of gastric carcinoma.KISS-1 and MMP-9 could be the markers to evaluate invasion and metastasis in gastric carcinoma.
