白藜芦醇对小鼠脑组织线粒体形成的诱导作用观察 被引量:1

摘要 目的观察白藜芦醇(Resveratrol,Res)对脑组织线粒体形成的影响,探讨其影响线粒体形成的可能机制。方法实验动物被随机分为3组,分别为空白对照组、羟甲基纤维素组和Res组,15天后检测脑组织中ATP含量和mtDNA表达水平,电镜观察线粒体变化,采用蛋白印迹和Real-time PCR法检测线粒体形成相关分子表达变化。结果 Res能够明显促进脑组织中ATP含量的增加,提高mtDNA表达水平,增加线粒体的数量。且蛋白印迹和Real-time PCR检测发现,Res能促进线粒体形成相关分子基因和蛋白的表达。结论 Res预处理可能具有通过影响线粒体形成相关基因的表达来促进脑组织线粒体形成的潜能。 Purpose To investigate the effect of resveratrol on the brain mitochondrial biogenesis and the potential mechanisms.Methods Mice were divided randomly into sham group,carboxy methyl cellulose group and resveratrol group(50 mg/kg).After 15 days,the ATP content and mtDNA level were measured,the mitochondrial changes were observed by transmission electron microscopy,and the molecular signaling of mitochondrial biogenesis was evaluated by real-time PCR and Western blot.Results Compared with the other groups,the resveratrol group significantly improved the ATP content and mtDNA levels,increased the number of mitochondria.In addition,the expression of mRNA and protein of molecules related to mitochondrial biogenesis were enhanced.Conclusion The study suggests that resveratrol pretreatment might promote the brain tissue mitochondria biogenesis through affecting molecules related to mitochondrial biogenesis.
