不同盐度和Fe^(3+)浓度对小球藻生长、硝酸还原酶活性及基因表达的影响 被引量:7

摘要 论文以一株紫外诱变获得的蛋白核小球藻F-9-3为材料,研究了4个盐度和5个Fe3+浓度对其生长和硝酸还原酶(NR)活性及其基因表达的影响。盐度实验结果表明在0~0.30 mol L-1NaCl浓度范围内,小球藻生长较快;NR酶活性及其基因表达量都是在0.15和0.30 mol L-1NaCl较高。铁浓度实验结果表明在0.03和0.06 mmol L-1Fe3+浓度培养小球藻生长较快,NR酶活性和基因表达量较高;而高铁浓度组(0.12 mmol L-1)其生长受抑制,缺铁情况下nr基因表达量最高。因此适合该小球藻生长的最适盐度范围是0~0.30 mol L-1NaCl和0.03~0.06 mmol The effects of four different salinity and five iron concentrations on the growth, nitrate reductase (NR) activity and its expression were investigated in Chlorella pyrenoidosa F-9-3, which was selected after ultraviolet radiation. The salinity experiment showed that C. pyrenoidosa F-9-3 grew faster in 0-0.30mol-LqNaCl medium than in 0.45 mol.LqNaC1 medium. And NR activity and its expression were higher in 0.15 and 0.30mol.LlNaCl groups than those of other two groups. The iron experiment showed that the growth, NR activity and gene expression of C. pyrenoidosa F-9-3 were all higher in 0.03 and 0.06 mmol.L'lFe3+ treatments than those in other Fe3+ concentration mediums. The growth was inhibited in 0.12 mmol.L-1 iron medium and NR expression was the highest in iron deficiency group. It can be concluded that optimum culture condition of the alga was 0-0.30 mol.LlNaCl and 0.03-0.06 mmol-Lq iron concentration.
