摘要
目的建立HPLC法同时测定叶下珠中5种成分没食子酸、原儿茶醛、柯里拉京、鞣花酸、槲皮素的含量。方法采用RP-HPLC测定叶下珠中5种成分的含量。采用Alltima C18(4.6×150mm,5μm),流动相为甲醇-0.1%磷酸梯度洗脱,流速为1.0mL/min,检测波长为280nm,柱温为30℃,进样体积为10μL。结果没食子酸、原儿茶醛、柯里拉京、鞣花酸、槲皮素线性范围分别为0.198~3.166μg(r=0.9998)、0.195~3.128μg(r=0.9996)、0.235~3.754μg(r=0.9999)、0.140~2.234μg(r=0.9997)、0.179~2.862μg(r=0.9995);平均加样回收率分别为99.82%(RSD=2.48%)、99.68%(RSD=1.48%)、100.43%(RSD=1.49%)、95.21%(RSD=2.42%)、100.34%(RSD=2.27%)。结论本研究所建立的方法方便、结果稳定可靠,可用于叶下珠的质量控制。
Objective To establish a HPLC method to simultaneously determine five active components including gallic acid,corilagin,ellagic acid,protocatechuic aldehyde and quercetin in Phyllanthus amarus L.Methods The contents of five active components in Phyllanthus amarus L was determined by HPLC.An Alltima C18(4.6×150mm,5μm) column was used for sample analysis of Phyllanthus amarus L..Methanol-0.1% phosphoric acid was used for the mobile phase by a 1.0mL/min gradient elution.The 280nm wavelength was used for detection of samples.The column temperature was kept at 30℃ and injection volume was 10μL.Results The five compounds all had a good linear relationship.The linear range of gallic acid,protocatechuic aldehyde,corilagin,ellagic acid and quercetin were 0.198-3.166μg(r=0.9998),0.19-3.128μg(r=0.9996),0.235-3.754μg(r=0.9999),0.140-2.234μg(r=0.9997),0.179-2.862μg(r=0.9995).The average recovery rate were 99.82%(RSD=2.48%),99.68%(RSD=1.48%),100.43%(RSD=1.49%),95.21%(RSD=2.42%) and 100.34%(RSD=2.27%).Conclusion The established HPLC method is a convenient,accurate and can be used for the quality control of Phyllanthus amarus L.
