摘要
目的:探讨刺五加皂甙(Acanthopanax Senticousus Saponins,ASS)对胚基底前脑乙酰胆碱酯酶(acetyl-cholinesterase,AChE)阳性神经元发育的影响。方法:将孕17 d SD大鼠胚基底前脑原基制成细胞悬液接种于24孔培养板中。分别加入含12.5、25、50、75 mg/L的ASS(ASS组),或100 ng/mL的神经生长因子(nerve growth factor,NGF组),及不含上述成份(对照组)的DMEM培养液,于体外培养18 d后,进行AChE组织化学染色,显微镜下统计各组每孔中的AChE阳性神经元数,并用CMM-301图像分析系统对AChE阳性神经元的细胞面积和细胞周径进行统计,数据用单因素方差分析进行统计学处理。结果:ASS各质量浓度组AChE阳性神经元数量均显著多于对照组(P<0.01);ASS各质量浓度组AChE阳性神经元的面积和周径的指标亦明显好于对照组(P<0.01)。结论:ASS可能通过促进胚基底前脑AChE阳性神经元的发育来改善认知功能。
Objective: To study the effects of Acanthopanax Senticousus Saponins(ASS) on the development of AChE positive neurons in the embryonic basal forebrain.Methods: The cell suspension was prepared from embryonic basal forebrain of E17-day old SD rats.The cell suspension was placed into 24-well cultured plates which had DMEM medium containing ASS of 12.5 mg/L,25 mg/L,50 mg/L and 75 mg/L(ASS group) and NGF of 100 ng/mL(NGF group) and not containing the above elements(control groups).After 18 days of culturing,the cultured neurons in the 24-well plates were detected by using AChE histochemical stains.The number of AChE positive neurons was counted under the microscope,and the cell areas and circumferences of these neurons were analysed with CMM-301 computer image system.The data were statistically treated with the analysis of variance(ANOVA) and the test of Turkey.Results: The number of AChE positive neurons in the ASS groups of each consistence was apparently greater than that in the control group(P0.01).The cell areas and circumferences of AChE positive neurons in the ASS groups of each consistence were also much superior to those in the control group(P0.01).Conclusions: Acanthopanax Senticousus Saponins can promote the development of AChE positive neurons in the embryonic basal forebrain,improve cognition.
