摘要
目的检测汉坦病毒感染人淋巴细胞后人正常T细胞表达分泌调节活化因子(RNATES)和干扰素调节因子-7(IFR-7)表达情况,并为深入探讨可能其致病机制奠定基础。方法本研究用汉滩病毒感染人淋巴细胞系LLC,取感染后不同时间点的细胞提取总RNA,半定量RT-PCR检测感染细胞中IFR-7及RANTES mRNA的变化情况。结果在汉滩病毒感染人淋巴细胞早期,可诱导IRF-7和RNATES基因的转录,并随着感染时间的延长表达持续上调。结论汉坦病毒可以感染人淋巴细胞,并在感染早期持续上调RNATES和IRF-7表达,对深入研究汉坦病毒的致病机制有重要意义,也为寻找HFRS的治疗药物靶位奠定基础。
The pathogenesis of hemorrhagic fever with renal syndrome(HFRS) is not very clearly,but more and more studies indicate that the immunoreaction is one of most important factors.In this study,the total RNAs were extracted from LLC cells infected by 76-118 strain of Hantaan virus with different intervals and subjected to reverse transcription.Meanwhile,semi-quantitative RT-PCR was used to detect the mRNAs of RANTES and IRF-7.Finally,the PCR products amplified with RANTES and IRF-7 primers were cloned and sequenced.The results showed that RANTES and IRF-7 mRNAs were detectable in LLC infected by Hantaan virus,and the expression levels were increased as the infection time prolonging.It is concluded that Hantaan virus could infect LLC,and the expression of RANTES and IRF-7 could be upregulated in the early stage of the infection.The study is of great importance to analyze the pathogenesis of Hantavirus and the therapeutic approach of HFRS.
