摘要
在皱落假丝酵母SIPI2.230发酵时,如在培养基中加入少量吐温-80及脂类物质,可使脂酶发酵单位从原来的1.6u/ml提高到100u/ml。脂酶提纯时,采用DEAE-纤维素搅拌吸附与洗脱、Bulyl-Toyopearl650 M疏水层析及交联葡聚糖G-100凝胶过滤层析工艺,可提高酶纯度66.8倍。
In the preparation of lipase from Candida rugosa SIPI 2.230, itwas found that addition of a small amount of a surfactant Tween-80 and oilssuch as soybean oil, sesame oil, linseed oil or olive oil can increase the lipaseproduction from 1.6 to 100 u/ml. The enzyme was purified by batch-wise adsorption and elution on DEAE-cellulose, Butyl-Toyopearl 650M hydrophobicchromatography and Sephadex G-100 gel filtration chromatography, The purityof the enzyme was increased by 66.8-fold.
