通用引物单链构象多态性技术检测呼吸道常见病原菌的研究 被引量:2

Identification of pathogens responsible for respiratory tract infections using universal primer single-strand conformation polymorphism

摘要 目的探讨通用引物单链构象多态性技术用于呼吸道常见病原菌检测的可行性。方法选取7种常见呼吸道感染细菌,采用通用引物对细菌的16SrRNA基因进行扩增,对扩增产物进行单链构象多态性分析。结果经过扩增,7种常见病原菌均在300bp处有清晰条带,扩增产物的单链构象多态性图谱条带数为2~4条,相对迁移率及条带间距也存在明显不同,各细菌间可相互区分。结论通用引物SSCP技术能快速简便地鉴定呼吸道常见病原菌。 Objective To explore use of universal primer single-strand conformation polymorphism(SSCP) to identify pathogens responsible for respiratory tract infections.Methods The 16S ribosomal RNA genes of seven typical pathogens responsible for respiratory infections were amplified by universal primer polymerase chain reaction and the PCR products were analyzed for single-strand configuration polymorphism.Results According to PCR,seven pathogens responsible for respiratory infections had distinct bands of 300 bp and 2-4 bands in the SSCP profiles.Relative mobility and the distance between bands differed markedly,allowing identification of all of the pathogens.Conclusion Universal primer SSCP analysis can rapidly and conveniently identify pathogens responsible for respiratory tract infections.
