不同地理株旋毛虫感染小鼠IL-2水平变化及肌幼虫分布研究 被引量:1

Level of IL-2 and the distribution of muscle larvae in mice infected with different geographical isolates of Trichinella

摘要 目的观察河南株、云南株和黑龙江株旋毛虫感染小鼠后IL-2水平变化及肌幼虫分布的差异。方法取昆明株雄性小白鼠120只,体重18~22g,随机分成河南株、云南株和黑龙江株感染组,每组30只,每鼠分别感染相应旋毛虫幼虫200条。另设1个正常对照组。ELISA法检测小鼠感染旋毛虫后第1、2、3、4、5、6周血清IL-2含量;第6周处死小鼠,做肌肉压片,镜检,观察不同组感染鼠肌肉中的旋毛虫幼虫分布情况,比较不同组同一部位肌幼虫分布的差异。结果小鼠感染河南株、云南株和黑龙江株旋毛虫后血清IL-2分泌量均增加,其中感染后第1周达到峰值,第5周时下降至正常水平,第6周时IL-2含量略低于正常水平;不同地理株旋毛虫感染均可导致宿主肌幼虫数量的增加,但是不同虫株感染后小鼠肌幼虫数不同,以河南株感染鼠肌幼虫较多。结论不同地理株旋毛虫感染后小鼠血清IL-2变化趋势相同,同一时期河南株感染鼠IL-2含量最高,云南株感染鼠略低,黑龙江株感染鼠最低;感染鼠同一部位肌幼虫分布有所差异,感染度以河南株感染鼠最重,云南株感染鼠次之,黑龙江株感染鼠最轻。 Objective To investigate the levels of IL-2 and the distribution of muscle larvae in mice infected with Henan,Yunnan,and Heilongjiang isolates of Trichinella.Methods One hundred and twenty Kunming male mice(weight: 18 to 22 g) were grouped randomly into a group infected with a Henan isolate,a group infected with a Yunnan isolate,and a group infected with a Heilongjiang isolate(each were infected with 200 T.spiralis larvae) and a control group,with 30 mice in each group.ELISA was used to measure the level of IL-2 in serum from the first week to the sixth week after the mice were infected with Trichinella.The mice were sacrificed in the sixth week and the distribution of encapsulated muscle larvae was observed in each group.The distribution of muscle larvae at the same site was compared among the groups.Results The levels of IL-2 increased after the mice were infected with the Henan,Yunnan,and Heilongjiang isolates of Trichinella,and then they gradually returned to normal in the fifth week and were lower than normal in the sixth week.Different isolates resulted in an increase in the distribution of muscle larvae,but the number of muscle larvae differed in mice infected with different isolates.The group infected with the Henan isolate had the severest infection among the three groups.Conclusion The levels of IL-2 were the same in the three groups infected with different isolates of Trichinella.During the same time period,the highest levels of IL-2 were found in mice infected with the Henan isolate,the lowest levels of IL-2 were found in mice infected with the Heilongjiang isolate,and medium levels of IL-2 were found in mice infected with the Yunnan isolate.The number of muscle larvae in the same muscle differed in mice infected with different isolates.The group infected with the Henan isolate had a severe infection while the group infected with the Heilongjiang isolate had a mild infection.
