摘要
[目的]探讨ErbB-3结合蛋白Ebp1基因对肝癌细胞生长增殖的影响.[方法]利用脂质体法将pEGFPC1-Ebp1融合质粒转染到人肝癌细胞HepG2中,在荧光显微镜下观察绿色荧光蛋白融合基因的表达并建立稳定表达Ebp1基因细胞系,应用Western blot检测转染后目的蛋白的表达,利用平板克隆集落形成观察细胞生长增殖能力的改变.[结果]在荧光显微镜下可见绿色荧光,Ebp1融合蛋白主要表达于胞质,转染pEGFPC1-Ebp1质粒后HepG2细胞中蛋白明显呈高表达;克隆集落形成实验结果表明,pEGFPC1空载体转染组和未转染细胞组相比较,Ebp1过表达细胞克隆集落形成显著增多.[结论]Ebp1过表达可增强人肝癌HepG2细胞增殖.
OBJECTIVE To study the effect of ErbB-3 binding protein Ebpl gene on the growth and proliferation of the HepG2 hepatic carcinoma cells. METHODS The pEGFPC1-Ebpl fusion gene were transfected into the human hepatic carcinoma cells (HepG2), the expression of Ebpl was observed with fluorescence microscope and used for detecting the expression of Ebpl protein by Western blot; the effect of Ebpl on HepG2 cells were identified by detecting the cell cloning formation assay. RESULTS The detection of Western blot indicated that Ebpl protein was over expressed HepG2 ceils transfected by pEGFPC1-Ebpl. Colony formation assay indicated that the colony formation ability of Ebpl-HepG2 cells was enhanced. CONCLUSION That the ectopic expression of Ebpl effectively enhance the growth and proliferation of HepG2 hepatic carcinoma cells.
