续断对兔骨缺损修复中BMP-2基因表达的影响

Effects of the teasel root for BMP-2 gene expression in repairment of bone defect of rabbits

[目的]观察续断在兔桡骨骨缺损修复过程中对BMP-2基因表达的影响.[方法]取30只家兔随机分成5个组,在双侧桡骨中段制作20mm长骨缺损模型.取自体松质骨颗粒,植骨于骨缺损处.实验组用续断按照20g/kg剂量混合于饲料中喂养,对照组单纯喂养饲料.于术后1,2,4,6,8周行X线检查后取骨痂用免疫组织化学及Real Time RT-PCR方法检测BMP-2基因的表达变化.[结果]实验组1周时BMP-2呈弱阳性表达,2周时呈阳性表达,4周时呈强阳性表达,6周及8周时呈阳性表达.对照组4周时呈阳性表达外1,2,6,8周时均呈弱阳性表达.实验组1周时较对照组BMP-2基因表达量无明显差异,2,4,6,8周时实验组BMP-2基因表达量均较对照组明显增加(P<0.05),4周时表达量增加最明显.[结论]续断在骨缺损修复过程中有明显的增加BMP-2基因表达的作用,可促进骨缺损的修复.

OBJECTIVE To study the effects of the teasel root for BMP-2 gene expression in repairment of radius defect of rabbits. METHODS 20 mm length of bone defect models were made in both sides of radius of 30 rabbits, and the autologous cancellous bone was grafted to the place of radius defect. The rabbits in experimental group were fed with feed with mixing the teasel root by 20 g/kg, and in control group were fed with normal feed. After 1, 2, 4, 6 and 8 weeks and X-ray examination, new formed bone was taken to detect the BMP-2 gene expression by immunohistochemistry and real-time RT-PCR. RESULTS In experimental group, BMP-2 expression was weakly positive at a week, positive at 2 weeks, strong positive at 4 weeks, and positive at 6 weeks and 8 weeks, and in control group, BMP-2 expression was positive at 4 weeks, and weakly positive at 2, 4, 6, 8 weeks. BMP-2 gene expression at a week was no significant difference between two groups, but BMP-2 gene expression at 2, 4, 6, 8 weeks was markedly increased in experimental group than in control group（P〈0.05）, especially at 4 weeks. CONCLUSION The teasel root significantly increase the expression of BMP-2 gene in repairing process of bone defect, and can promote the repairment of bone defect.
