响应面法优化植物乳杆菌R23产苹果酸乳酸酶条件 被引量:1

Optimization on Producing Malolactic Enzyme Condition of Lactobacillus Plantarum R23 Using Response Surface Methodology

摘要 为获得高产量的苹果酸乳酸酶(MLE),以植物乳杆菌R23为发酵菌株,在前期单因素试验的基础上,采用响应面设计(RSD),建立二次多项数学模型,优化植物乳杆菌R23的产MLE条件。试验结果表明,对植物乳杆菌R23产MLE的总活力的影响次序是:起始L-苹果酸浓度>发酵温度>起始pH值;植物乳杆菌R23产MLE的最佳培养条件是:发酵温度33.5℃、pH值6.2、L-苹果酸质量浓度3.8g/L,在此条件下植物乳杆菌R23厌氧发酵28h的产酶量为1.34mg/mL,酶活力462.33u,总活力619.52U。 In order to enhance production of malolatic enzyme(MLE) of Lactobacillus plantarum R23, response sur- face methodology was used to optimize condition of MLE production on the basis of pre-experiments, and a second order quadratic equation was finally obtained. Results showed that the effect order of three factors to the total acticty of MLE is as follows: concentration of L-malic acid〉fermentation temperature〉pH value. The optimized conditions were fermenta- tion temperature 33.5℃, pH value 6.2, concentration of L-malic acid 3.8 g/L. Under the optimal conditions, MLE pro- duction of Lactobacillus plantarum R23 with anaerobic fermentation 28 h was 1.34 mg/mL, MLE activity and the total ac- tivity was 462.33 u and 619.52 U respectively.
