摘要 目的:探讨microRNA-1(miR-1)对施万细胞增殖的影响及其作用的靶基因。方法:采用Edu染色法检测miR-1对原代培养的施万细胞增殖能力的影响,用双荧光素酶报告基因系统确定miR-1的靶基因。结果:miR-1能够抑制原代培养的施万细胞的增殖,并可直接作用于Edn1基因的3′UTR。结论:miR-1可以直接靶向Edn1的3′UTR,从而通过下调靶基因Edn1的表达抑制施万细胞的增殖。 Objective:To investigate the influence of miR-1 on the proliferation of Schwann cells and the target gene of miR-1.Methods: Edu staining analysis and dual luciferase reporter system were used to examine the effect of miR-1 on proliferation ability of primary cultured Schwann cells and to identify the target gene of miR-1,respectively.Results: MiR-1 inhibits the proliferation of primary cultured Schwann cell proliferation and targets the 3’UTR of Edn1 gene directly.Conclusion: This study reveals that miR-1 targets 3’UTR of End1 gene directly and inhibits the proliferation of Schwann cells by down-regulating the expression of its target gene Edn1.
