吴茱萸汤对虚寒型偏头痛小鼠单胺类神经递质及一氧化氮含量的调控作用

The Regulative Effects of Wuzhuyu Decoction on the Content of Monoamine Neurotransmitters and NO in Model of Migraine of Deficiency-Cold Syndrome in Mice

目的:建立利血平化低5-羟色胺(5-Hydroxytryptamine,5-HT)伴局部脑血管痉挛型小鼠偏头痛模型,观察复方吴茱萸汤对该模型的作用。方法:采用皮下注射利血平(0.13 mg.kg-1)并大脑皮层埋设自体血凝块(2μL.只-1)方法制备小鼠偏头痛模型。按小鼠体质量随机分为正常对照组、假手术组、模型组、吴茱萸汤组和阳性药组。造模同时吴茱萸汤组及阳性药组分别给予吴茱萸汤(29.4 g生药.kg-1)及琥珀酸舒马普坦(7.5 mg.kg-1)治疗,假手术组、模型组及正常对照组给予等剂量生理盐水灌胃,观察药物对小鼠脑组织5-HT等单胺递质及脑组织、血浆中一氧化氮(Nitric-oxide,NO)含量的影响。结果:造模后模型组小鼠脑组织内5-HT、DA及NE等单胺类神经递质含量与正常组比较,差异有统计学意义(P<0.01);血清中NO含量与正常组比较,差异有统计学意义(P<0.05);脑组织内NO含量与正常组比较,差异有统计学意义(P<0.05)。吴茱萸汤能升高模型鼠脑组织内5-HT、DA,血清内NO含量,降低脑组织内NO含量,与模型组比较,差异均有统计学意义(P<0.05),而阳性药琥珀酸舒马普坦仅能升高模型鼠脑组织内5-HT(P<0.05)。结论:本实验采用的造模方法稳定,可行;吴茱萸汤对上述指标均有改善作用,阳性药作用单一。

Objective：To establish a mice model of migraine marked by low 5-Hydroxytryptamine induced by reserpine with local cerebral vasospasm,then observe the effects of compound Wuzhuyu decoction on the model.Methods：The mice migraine model was induced by reserpine via subcutaneous injection and the blood clots injected into cerebral cortices（2 μL）.At the same time Wuzhuyu-decoction group and positive control group were given Wuzhuyu-decoction（29.4 g·kg-1） and sumatriptan succinate（7.5 mg·kg-1） respectively for treatment.To observe the different effect of that two kind of drugs on the content of neurotransmitters such as 5-HT in brain tissues and NO（nitric oxide） in brain tissues and blood serum.Results：In the modeling group,the content of 5-HT,DA and NE etc.monoamine neurotransmitters in brain tissues are significantly reduced（P0.01）,the concentration of NO in blood serum is significantly reduced while that in the brain tissues is significantly increased（P0.05）.Wuzhuyu decoction can improve the decrease of 5-HT and DA in the brain tissues,increase the concentration of NO in the serum,reduce that in brain tissues,and there are significantly differences comparing with the model group（P0.05）.Conclusion：The experimental method of the model is stable and feasible;Wuzhuyu decoction has an effect to improve the indexes we obtained while the target of positive control drug is mere 5-HT.
