载药骨基质明胶治疗骨髓炎的实验研究

Experimental study in the treatment of osteomyelitis with cefazolin loaded bone matrix gelatin

目的 :探讨载药骨基质明胶 (BMG)治疗骨髓炎的可能性。方法 :将 BMG溶于头孢唑林钠溶液 ,通过真空吸附、冻干等处理 ,制得载药 BMG。测定其体内、体外释药浓度及持续时间以及载药 BMG诱导成骨的能力。结果 :载药BMG对金葡菌的抑制作用体外为 2 2 d,体内为 14d。载药 BMG体内释药时 ,局部组织浓度高 ,血浆浓度低 ;局部组织早期浓度高 ,以后为稳定的低浓度释放。载药 BMG中掺入抗生素后不影响 BMG的诱导成骨能力。结论 :载药 BMG既能持续释放有效浓度的抗生素 ,又具有较强的诱导成骨能力 。

Objective:[WT5”BZ]To investigate the possibility of Cefazolin loaded bone matrix gelatin (C BMG) used for the treatment of osteomyelitis Methods:BMG was dissolved in cefazolin solution,then absorbed and lyophilized in vacuum,and prepared as C BMG,In vivo and in vitro drug delivery analyses of C BMG were performed The cefazolin concentration of experimental samples was determinted by HPLC assay The osteoinductive ability of C BMG was also assessed Results:A high concentration of cefazolin released from BMG sustained for 22 days in vitro and 14 days in vivo The cefazolin concentrations were lower in plasma than those in local bone tissues In the local bone tissues,cefazolin could be released in a high concentration in the early period,and the in a steady,low concentration In addition,cefazolin in the C BMG did no affect the osteoinductive ability of C BMG Conclusion:C BMG may become an ideal and new carrier and delivery system for treating osteomyelitis
