微波连续化辅助酶解大豆分离蛋白的工艺研究 被引量:4

摘要 研究了微波连续条件下,碱性蛋白酶水解大豆分离蛋白的工艺。通过单因素和正交设计试验,得到的最佳工艺条件为,微波功率500 W、底物浓度4%,酶底物比9%,酶解时间15 min,在此条件下水解度为27.33%,多肽含量为13.04 mg/mL,水浴对照实验发现微波连续辅助酶解15 min和普通水浴酶解1.5 h相当。电泳结果表明,酶解物分子量在20 000 u以下。 With microwave-assisted continuous technique,soybean protein isolates were hydrolyzed by alkaline protease.Through single factor and orthogonal design experiments,the optimal microwave-assisted alkaline protease hydrolysis condition for soybean protein isolates was microwave power 500 W,substrate concentration 4 %,the ratio of enzyme and substrate 9 %,reaction time 15 min,and the content of peptide was 13.04 mg/mL.Water bath controlled trial found that peptides hydrolyzed for 15 min under continuous microwave had the same result with those for 1.5 h under water bath.Gel electrophoresis showed that molecular weight of hydrolysates was less than 20 000 u.
