摘要
对灰白青霉HML0233进行紫外诱变,从中出筛选出比出发菌株FPA酶活提高了66.6%的突变菌株HBQ14,酶活提高到1.46×10-7kat/mL,经过发酵条件初步优化研究,以NH4NO3为氮源,40℃,pH5.5培养4 d,FPA酶活达到2.35×10-7kat/mL,灰白青霉HBQ14遗传性状稳定。
Ultraviole(UV)was used to induce mutagenesis on Penicillium canescens HML0233 and to select out a new strain highly producing cellulase.A new P.canescens strain HBQ14 with high yield of cellulase was obtained with the enzyme production amount 1.66 times over that of starting strain.This strain showed highest cellulase yield under the culture condition of 40 ℃ for 4 d,the nitrogen source was NH4NO3,pH 5.5,the enzyme activity was increased from 1.46×10-7 kat/mL to 2.35×10-7 kat/mL.The strain HBQ14 was also inherited stably after 8 generations.
