摘要
目的 研究大鼠大离中动脉闭塞 (MCAO)后同侧黑质的变化。方法 35只SD大鼠 ,用Tamura法作脑缺血模型 ,分别对缺血后 2周、1个月、3个月、6个月的鼠脑切片用微管相关蛋白 - 2作免疫组化和甲紫醇染色。用计算机测定黑质长度与面积 ,用计数器计算神经细胞数。结果 用 10 0 %表示对侧黑质的长度。同侧黑质长度 ,MCAO 2周时为 84% ,1个月 81% ,3个月 77% ,6个月 6 4%。用 1表示对侧黑质的面积 ,MCAO侧黑质的面积 2周时为 0 .83,1个月 0 .6 6 ,3个月 0 .5 2 ,6个月 0 .48。MCAO侧黑质神经细胞与对侧黑质神经细胞数的比 ,MCAO2周时下降到 80 % ,1个月 6 8% ,3个月 5 3% ,6个月 46 %。结论 MCAO后黑质的长度缩短 (P <0 .0 1)、面积缩小(P <0 .0 0 1) ,同时神经细胞数也减少 (P <0 .0 0 1)。
Objective To investigate the change of ipsilateral substantial nigra after middle cerebral artery occlusion(MCAO).Methods Thirty-five SD rats were operated by Tamura's method,brain slices of rats were immunohistochemically stained with microtuble-associated protein-2 (MAP-2) and Luxol fast blue-Cresyl violet 2 weeks after MCAO,1 month,3 months,6months.The length and areas of substantia nigra were measured by computer,neuronal cells were counted by counter.Results The length of contralateral substantia nigra of ischemia was taken as 100%,so the length of ispilateral substantia of ischemia was 84%,2 weeks after MCAO,1 month 81%,3 months 77% and 6 months 64%.1 was used to exspress the areas of no-ischemic lateral substantia nigra,so the areas of ispilateral substantia nigra were 0.83 2 weeks after MCAO,1 month 0.66,3 months 0.52 and 6 months 0.48.Neuronal cells in ischemia ipsilateral substantia nigra were decreased to 80% 2 weeks after MCAO,1 month 68%,3 months 53% and 6 months 46%,compared with contralateral substantia nigra.Conclusion The length and areas of ipsilateral substantia nigra are reduced and neuronal cells are decreased significantly, P<0.01,P <0.001 and P<0.001 respectively.
