HPLC-ELSD法测定景天祛斑胶囊中黄芪甲苷的含量

Determination of AstragalosideⅣ in Jingtianquban Capsules by HPLC-ELSD

目的建立景天胶囊中黄芪甲苷的含量测定方法。方法采用HPLC-ELSD法,色谱柱为ZORBAX SB-C18(5μm,250mm×4.6mm);流动相为乙腈-水(32∶68);流速为0.8min·mL-1,柱温为30℃。PL-ELS2100型蒸发光散射检测器(漂移管温度:80℃,雾化温度:50℃),载气(氮气)流速1.5mL·min-1。采用外标两点法计算含量。结果黄芪甲苷在选定浓度0.00525～0.0525mg·mL-1内有良好的线性关系(r=0.9959),回收率为100.85%(RSD=1.73%)。结论本法方便、快速、准确,重现性好,可用于该制剂中黄芪甲苷的定量分析。

OBJECTIVE To establish a method to determine astragaloside Ⅳ in Jingtianquban Capsules.METHODS HPLC-ELSD was used to directly determine astragaloside Ⅳ on ZORBAX SB-C18（5μm,250mm×4.6mm） column,Using acetonitrile-water（32∶68） as mobile phase,the flow rate was 0.8mL·min-1.The column temperature is at 30℃.Detector：PL-ELS2100 ELSD（Eva：80℃,Ned：50℃）.The flow rate of gas was 1.5mL·min-1.Calculate contents with external standard two-point method.RESULTS AstragalosideⅣshowed a good linearity in the range of 0.00525～0.0525mg·mL-1,r=0.9959,the average recovery was 100.85%（RSD=1.73%）.CONCLUSION The method is simple,rapid,accurate and with good reproducibility.It can be applied in quantitative determination of astragaloside Ⅳ in Jingtian Capsules.
