雌三醇及三苯氧胺对人结肠癌细胞内游离钙离子浓度的影响

摘要 目的 :探讨雌三醇 (E3 )及三苯氧胺 (TAM )对人结肠癌细胞内游离钙离子 ( [Ca2 + ]i)水平的影响及其意义。方法 :采用流式细胞仪动态检测E3 及TAM作用后 ,人结肠癌细胞内 [Ca2 + ]i水平的变化。结果 :①E3 使细胞内 [Ca2 + ]i水平下降 ,并维持在一个较低水平 ( 2 7 88nM ) ,与静息状态细胞内 [Ca2 + ]i水平 ( 94 40nM )相比有显著性差异 (P <0 0 5 )。②TAM使细胞内 [Ca2 + ]i水平上升 ,并维持在一个较高水平 ( 3 4 9 97nM ) ,与静息状态细胞内 [Ca2 + ]i水平( 10 7 88nM)相比有显著性差异 (P <0 0 5 )。结论 :①E3 不引起胞内 [Ca2 + ]i水平升高 ,E3 促人结肠癌细胞增殖作用的机理符合甾体激素的基因调节作用 ,未见与钙离子通道的非基因的快速调节作用有关。②TAM抑制人结肠癌细胞增殖的机理 ,除通过竞争性结合雌激素受体‘外 ,还可能通过了改变胞内 [Ca2 + ]i浓度等途径。 Object:To study the effect of estriol(E 3)and tamoxifen (TAM)on the concentration of intracellular free,calcium in human colonic cancer cells and analyses the significance of the effect.Methods:Flow cytometry was used to continuously monitor the change of the intracellular free calcium([Ca 2+ ]i) in human colonic cancer cells(HT 29 cells )treated with E 3 and TAM.Results:First,E 3 made the intracellular [Ca 2+ ]i concentrations maintained a lower level (27 88nM)and was much lower than that of the intracellular [Ca 2+ ]i of static status cells(94 4nM).There was statistically difference between 27 88nM and 94 4nM( P <0 05).Secondly,TAM made the intracellular [Ca 2+ ]i level increase.Then the intracellular [Ca 2+ ]i level maintained a higher level (349 97nM)and was much higher than that of the intracellular[Ca 2+ ]i of static status cells(107 88nM).There was statistically difference between 349 97 nM and 107.88nM( P <0 05).Conclusion:① E 3 can not cause the intracellular [Ca 2+ ]i increase.The effect mechanism of E 3 due to the effect of gene regulation,but not the effect of rapid non gene regulation by Ca 2+ cannel.② TAM can inhibit the proliferation of cancer cells by changing the intracellular [Ca 2+ ]i of concentration and competitively combine the estrogen receptor.
