摘要
目的 探讨流式细胞术CD45辅助设门在白血病表型分析中的作用。方法 根据CD45在所有白细胞上都有表达 ,但在不同系成熟细胞或同系不同发育阶段细胞表达量不同的特点 ,在流式细胞仪上采用对数CD45 -线性侧向角光散射 (SSC)图设门及前向角光散射 (FSC) -侧向角光散射 (SSC)图设门进行免疫表型分析。结果 用FSC -SSC表示则白血病细胞群与正常淋巴细胞群分布领域的境界不清 ,而用CD45 -SSC图表示可把两群细胞明显区分开来 ,且两种设门方法所得表型结果有不同之处。结论 CD45 -SSC图设门提高了白血病免疫学分型的准确性 ,尤其对白血病细胞比例较低患者提供了一个可靠的方法学手段 ,且该方法易于掌握 ,无主观因素 ,重复性好。
Objective To approach into the function of CD45 auxiliary gating in analysing the leukemic cell immunophenotype by flow cytometry. Method Basing on the expression of on all leukocytes, but with different amounts in cells of separate lineages and cells of a single lineage at various stages of maturation, CD45-SSC gating and FSC-SSC gating on flow cytometric device were used to analyse the immunophenotype of cells. Results Expression on FSC-SSC gating, here was no clear demarkation between leukemic cells and normal lymphatic cells, while on CD45-SSC gating the two clusters of cells could beseparated and the phenotypes from the two methods were different. Conclusion CD45-SSC gating can increase the accuracy in immunoloqical classification of leukemia especially in patients with relatively low count of leukemic cells. It provides a reliable technological method, easy to master, with high reproducibility and without subjective factors.
