摘要
目的建立测定K562/G01细胞株(imatinib-resistant K562 cells,耐伊马替尼K562细胞株)内伊马替尼浓度的方法,探讨甲基莲心碱对其细胞内STI571转运的影响。方法采用反相高效液相色谱法(reversedphase high-performance liquid chromatographic,RP-HPLC)直接测定药物浓度。色谱柱为:Gemini C18(250 mm×4.6 mm,5μm);流动相为:0.02 mol L-1KH2PO4-CH3CN=65:35;流速:1.0 mL min-1;检测波长:265 nm。结果采用外标法以峰面积定量,以色谱峰面积对STI571浓度进行线性回归,得回归方程:A=83.08C-7.975,r=0.999 4。线性范围49.51~3 076.92 ng mL-1,相对回收率分别>95%,日内日间RSD均<10%。结论该方法准确、快速、操作简便,可用于细胞内药物浓度的测定与研究。结果显示甲基莲心碱能影响细胞内STI571的转运。
Objective To develop an RP-HPLC method to detemine the imatinib concentration in K562/G01 cells(imatinib-resistant K562 cells),and to determine the effect of neferine on STI571 transportion.Methods RP-HPLC was used to determine the concentration of STI571.Assay was conducted using a Gemini C18(250 mm×4.6 mm,5 μm) with mobile phase consisting of 0.02 mol · L-1 potassium dihydrogen phosphate and acetonitrile(65 : 35) at a ·ow rate of 1.0 mL · min-1 and detected at 265 nm.Results Linear regression was made on the concentration of STI571 with peak area quantitate and chromatographic peak area the external reference method,the regression equation: A= 83.08C-7.975,r = 0.999 4.The relative recoveries were95%,the inter-and intra-day RSD was10%.Calibration plots in K562/G01 cells were linear at 49.51-3 076.92 ng · mL-1.Conclusion This method is accurate,quick and simple,which can be used for the determination of the intracellular concentrations of drugs.Neferine can affect the transportation of STI571.
