摘要
目的:选取四逆汤为经方代表,采用3种经方剂量折算标准,通过动物实验比较不同折算标准的药效及安全性。方法:复制大鼠失血性低血压模型,分成模型组和四逆汤高、中、低剂量组,取血清进行动脉血气分析,测定氧分压(PaO2)、二氧化碳分压(PaCO2)、剩余碱(BE)、碳酸氢根(HCO3-),生化仪检测血清中丙氨酸转氨酶(ALT)、天门冬氨酸转氨酶(AST)、尿素氮(BUN)、肌酐(Cr)、肌酸激酶(CK),乳酸(LA)、乳酸脱氢酶(LDH)的含量;取心、肾组织检测超氧化物歧化酶(SOD)和丙二醛(MDA)含量;取肺脏检测一氧化氮(NO)和一氧化氮合酶(NOS)含量。结果:与模型组相比,四逆汤低剂量和高剂量组均能显著降低PaCO2、升高(PaO2)(P<0.05,P<0.01);四逆汤低剂量组能降低LDH、LA、Cr、CK、ALT、AST水平(P<0.05,P<0.01),增加肾组织SOD含量和减少MDA含量(P<0.01),降低肺组织NO和T-NOS含量(P<0.05);四逆汤高剂量组能降低BE和cHCO3-(P)(P<0.05,P<0.01),减少心肌组织SOD含量和增加MDA含量(P<0.01)。结论:四逆汤在一定剂量下能改善失血性低血压大鼠缺血缺氧状态和心肺肝肾等重要脏器功能,提示在临床运用四逆汤时若增大各组分剂量,应对其可能出现的毒副作用予以重视。
Objective: To compare the pharmacologic effects and potential toxic effects of Sini Decoction(SND)with dosages of different conversion criteria.Methods: Forty male Wistar rats were randomly divided into Control group,SND L,M and H group.The changes of the arterial blood-gas levels,biochemistry,LA,LDH,MDA,SOD,NO and NOS were observed.Results: In SND L group,the levels of PaCO2,LDH,LA,Cr,CK,ALT,AST,MDA,NO and T-NOS decreased dramatically(P0.05,P0.01),the level of PaO2,SOD increased significantly(P0.05,P0.01).In SND H group,the levels of PaCO2,SOD,BE and cHCO3-(P) dropped obviously(P0.01),the level of PaO2and MDA increased markedly(P0.05,P0.01).Conclusion: SND with proper dosage appear to ameliorate hypoxia-ischemia status and protect and maintain the function of vital organs in bloodletting hypotension rats.The results suggested that we should pay more attention to the potential risk when increasing the dosage of SND.
