摘要
目的:观察左归丸对D-半乳糖致衰老大鼠血液抗氧化能力和淋巴细胞DNA损伤的影响。方法:2月龄雄性SD大鼠30只,随机分为青年对照组、衰老组、左归丸组,每组10只。衰老组每日腹腔注射D-半乳糖400mg/kg,连续60d;青年对照组每日腹腔注射等体积0.9%氯化钠溶液;左归丸组处理同衰老组,同时按每日相当生药量4.97g/kg灌胃给药。采用黄嘌呤氧化酶法和硫代巴比妥酸(TBA)法检测血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;采用单细胞凝胶电泳(SCGE)检测淋巴细胞DNA损伤情况。结果:与青年对照组相比,衰老组大鼠血清SOD活性下降(P<0.01)、MDA含量升高(P<0.01),淋巴细胞DNA损伤增加(P<0.01);给予左归丸能显著提高D-半乳糖致衰老大鼠血清抗氧化能力,减少淋巴细胞DNA损伤。结论:左归丸可通过提高血液抗氧化能力、减少淋巴细胞DNA损伤而发挥其延缓衰老的作用。
Objective: To investigate the effects of Zuogui Pill on blood anti-oxidative abilities and DNA damage of lymphocytes in aging rats induced by D-galactose.Methods: 30 male SD(2 months old) rats were equally divided into three groups randomly: young control group,old group,Zuogui Pill group.The rats in old group were administered D-galactose 400mg.kg-1.d-1 by intraperitoneal injection for 60 days consecutively and the rats in young control group were administered the equal volum normal sodium.And the intervention group were given Zuogui Pill with 4.97g.kg-1.d-1by intragastric administration at the same time.The SOD activity and MDA content in serum were measured.Single Cell Gel Electrophoresis(SCGE) was used to detect the DNA damage of lymphocytes.Results: Compared with young control group,the SOD activity were decreased and the content of MDA were increased in serum of the old group(P0.01).The DNA damage of lymphocytes was increased(P0.01).The variations above in aging rats were restored statistically in Zuogui Pill group.Conclusion: Zuogui Pill could slow down the aging progress through improving the blood anti-oxidative ability and decreasing the DNA damage of lymphocytes.
