光照节律改变对大鼠视网膜的影响及中药干预作用的研究 被引量:2

摘要 目的探讨光照节律改变对大鼠视网膜的影响及中药的干预作用。方法 SD大鼠45只随机分成3组,对照组为自然光环境,模型组与中药组饲养在光照控制箱内,光照明/暗交替时间为18 h/6 h,中药组予以中药灌胃,实验时间4个月,观察大鼠视觉电生理、视网膜组织学改变,并对视网膜过氧化物以及抗氧化酶活性进行分析。结果模型组最大混合反应(maximal combined response,Max-R)a、b波振幅下降54.3%、65.4%,明视视锥反应(cone response,Cone-R)a、b波振幅下降35.8%、38.5%,振荡电位(oscillatory potentials,OPs)OS2值、30Hz闪烁光N1-P1振幅下降,与对照组比较差异有统计学意义(P<0.05)。中药组与模型组相似,与对照组比较差异有统计学意义(P<0.05)。对照组光学显微镜下见视网膜各层分界清晰,排列整齐,模型组与中药组视网膜普遍萎缩变薄,层次不清。对照组视网膜丙二醛(malondialdehyde,MDA)含量低,超氧化物歧化酶(superoxide dismutase,SOD)活性高,与模型组及中药组比较差异有统计学意义(P<0.05)。中药组SOD活性比模型组高,差异有统计学意义(P<0.05)。结论光照节律改变对大鼠视网膜结构功能有一定影响,中药可能对视网膜功能有一定的保护作用,但作用位点及机制还需进一步研究。 OBJECTIVE To investigate the effect of the light rhythm on the retinal function,structure and the traditional Chinese medicine intervention function on experimental rats.METHODS Forty-five SD rats were divided into three groups randomly.The control group was exposed to natural light.The model group and traditional Chinese medicine groups were exposed to artificial light(light:dark=18:6h).The medicine group was given traditional Chinese medicine to fill the stomach.Four months later,F-ERG,retina histo-pathology inspection,estrogen content were measured to every group.RESULTS The amplitude of wave-a and wave-b of Max-R in model group decreased by 54.3% and 65.4% respectively.The amplitude of wave-a and wave-b of Cone-R in model group decreased by 35.8% and 38.5% respectively.The amplitude of OS of Ops and N1-P1 of 30Hz Flicker in model group decreased too.The amplitude of ERG in the model group was lower than that of the control group(P0.05).The ERG of the traditional Chinese medicine group was similar to that of the model group.The entire retina atrophied by optical microscope in the model group and traditional Chinese medicine group.The layers of retina were disordered in the model group and traditional Chinese medicine group.Retina was normal in the control group.The content of MDA in control group was lower than that of the model group and traditional Chinese medicine group(P0.05).The activity of SOD in control group was higher than that of the model group and traditional Chinese medicine group(P0.05).The activity of SOD in the traditional Chinese medicine group was higher than that of the model group(P0.05).CONCLUSIONS The abnormal light rhythm could affect the retinal function and structure.Traditional Chinese medicine maybe protect retina to a certain extent.But the mechanism of traditional Chinese medicine needs further discussion.
