和解聚散方对单侧输尿管梗阻大鼠肾间质纤维化的防治作用 被引量:22

The Preventive and Therapeutic Effect of Hejiejusan Formula in UUO-induced Renal Interstitial Fibrosis Rats

摘要 目的:研究和解聚散方对单侧输尿管梗阻(unilateral ureteral obstruction,UUO)大鼠肾间质纤维化的防治作用。方法:采用UUO建立大鼠肾间质纤维化模型,随机分为模型组、贝那普利组和和解聚散方组,另设假手术组;术后7d、14d、21d分批取材,检测血清肌酐、尿素氮,采用HE和Masson染色检测肾脏病理、免疫组化法检测肾组织TGF-β1的表达。结果:与模型组相比,和解聚散方组血清尿素氮、血清肌酐水平均降低(P<0.05),肾小管损害百分比和肾间质损害分值较低(P<0.05),肾组织TGF-β1表达减少(P<0.05)。结论:和解聚散方可以降低尿素氮、血清肌酐水平,减轻大鼠肾间质纤维化的程度,下调UUO大鼠肾组织TGF-β1的表达,验证了和解聚散法的正确性。 Objective:To study preventive and therapeutic effect of hejiejusan formula in unilateral ureteral obstruction (UUO) - induced renal interstitial fibrosis(RIF) rats. Methods: RIF model were established by UUO. All the UUO rats were randomly divided into model group, benazepril group and hejiejusan formula group. Healthy rats were chosen as the sham operation group. The rats were killed on the seventh, fourteenth and twenty - one days in batehes respectively after UUO. And then, measure the levels of serum creatinine and blood urea nitrogen, and use Hematoxylin - eosin staining and Masson staining to measure renal pathology. The protein expression of TGF - β1 in kidney tissues were detected by Immunohistoche -mistry. Results:Compared with model group, the levels of serum creatinine and blood urea nitrogen in hejiejusan formula groups were all significantly decreased( P 〈 0.05 ), tubular damage percentage and renal interstitial damage were all significantly decreased( P 〈0.05 ) ,the protein expression of TGF- β1 in hejiejusan formula groups were all significantly decreased( P 〈 0.05 ). Conclusion : Hejiejusan formula can reduce the blood urea nitrogen, serum creatinine level, and the extent of renal interstitial fibrosis, and it has the function of down -regulating the TGF -β1 expression in UUO rats. Besides,it has verified the correctness of the reconciliation and everything.
