摘要
目的探讨Notch1在小细胞肺癌多药耐药中的作用。方法首先通过QRT-PCR和Western-blot从基因和蛋白水平检测小细胞肺癌敏感细胞株(H69)和耐药细胞株(H69AR)中Notch1的差异表达;应用siRNA抑制耐药细胞株H69AR中的Notch1的表达,通过CCK8检测细胞对各种化疗(ADM,DDP,VP-16)的敏感性,流式细胞仪检测细胞凋亡率的变化。结果 Notch1在H69AR中的表达明显高于H69,抑制耐药细胞株中Notch1的表达能够增加细胞对化疗药物的敏感性,促进细胞的凋亡。结论 Notch1可能参与了调节小细胞肺癌对多药耐药性。
Objective To investigate the role of Notch1 on regulates muhi-drug resistance in small cell lung cancer. Methods Detected by Western blotting and real-time PCR(RT-PCR), differential expression of Notch1 gene and protein in both the drug- sensitive cell line H69 and ADM-resistant cell line H69AR. The Notch1 expression was blocked by the SiRNA in H69AR.Then the sensitivities of ceils to chemotherapy drugs such as ADM, DDP, VP-16 were detected by CCK8 assay, apoptosis rate by flow cytometry. Results The expression of Notch1 was significantly increased in H69AR cells than in the H69 cells. The sensitivities of H69AR cells to chemotherapy drugs were increased when down-regulated the expression of the Notch1,with the apoptosis were increased. Conclusion Notch1 may associate with chemotherapy resistance of small cell lung cancer.
